Somatic embryogenesis and genetic transformation of Thai cotton variety (SiSamrong 60)

Résumé : To produce Thai cotton varieties resistant to the most important insect #Helicoverpa armigera#, we set up a protocol of somatic embryogenesis and genetic transformation on the variety SiSamrong 60. Hypocotyl explants obtained from 7 day-old seedlings were plated on MS medium supplemented with different zeatin/NAA ratios. All explants plated gave calli with an efficiency of 100%. Calli obtained after one month were transferred on the same medium, on medium supplemented with growth regulator ratios inverted or hormone free medium (MSO). Those transferred from medium containing zeatin at 2 mg/l and NAA at 0,0l mg/l to MSO gave embryogenic calli at 50%. To promote the multiplication and germination of embryos, embrogenic calli were transferred on the same medium solidified by gelrite instead of agar or supplemented with different concentrations of glutamine. At 5 mM or 20 mM, glutamine permitted a fast embryogenic calli multiplication and transferred on MSO medium, the percentage of embryos developped into plantlets is much higher 33.20 to 37.60%. First experiments of gene transfer via #Agrobacterium tumefaciens# have been started. The synthetic gene encoding for #Bacillus thuringiensis# endotoxin was transferred to hypocotyl-explants. Two to three months later, first kanamycin resistant calli were obtained at 8.72% and 5.97% depending on #Agrobacterum# dilution using for the inoculation.
