Résumé : Microsatellites or simple sequence repeats (SSR) can be used for genomic mapping, population studies and DNA fingerprinting. We have identified and characterized SSR containing sites in the filamentous ascomycete Magnaporthe grisea, the causal agent of rice blast disease, by constructing and screening an enriched library and by searching public databases. Both methods proved useful in uncovering microsatellites. A total of 40 SSRs (25 from the database screen and 15 from the enriched library) were screened for product length polymorphism on six Magnaporthe grisea isolates including GUY 11 and 2539, the parents of a cross from which a genetic map of the fungus has been developed. The number of alleles at each site ranged from one to six when assayed on 3% agarose gels. Inheritance and linkage was determined for twenty polymorphic sites in 60 F1 progeny from the above cross. Analysis of allele segregation showed that all the markers segregated in the expected 1:1 ratio. Map positions were determined for all 20 with at least one marker mapping to each of the seven known chromosomes. The SSR markers were stable and easy to assay. A preliminary population study using the developed SSR markers on isolates from Thailand proved to be useful in helping to group and classify these individuals.
