Résumé : Detailed keypoint objectives of the first reporting period are: 1. Rational choice of defence genes to be introduced in European rice genotypes; 2. Construction of synthetic genes for AFP and CecA genes to be suitable for introduction into rice; 3. Preparation of transformation vectors for stable introduction of the defence genes driven by the strong constitutive promoter phobia; 4. Transformation activities for obtaining transgenic genotypes carrying the constructed genes; 5. Functional analysis of fungal/elicitor and wound/inducible promoters to be subsequently used for modulating gene expression in function of fungal attack; 6. Preparation of tools for verifying products. i.e. specific PCR primers, hybridisation probes and antisera; 7. Preparation of vectors for chloroplast targeting (plastomic transformation); 8. Evaluate the resistance to leaf blast, neck blast and stem rot at the level of young and mature plants of the parental genotypes; 9. Establish pathogenicity assays for transient expression studies of the interaction plant/pathogen (rice/Magnaporthe) in search for fungal/inducible promoters; 10. Construction of transgenic plants with the GUS marker gene under the proposed fungal-elicited promoter chosen, for studying the plant/pathogen interaction.
