Abstract : In situ hybridization of a Gmcal probe was performed in order to investigate whether the expression of this gene, encoding for a nodule specific carbonic anhydrase, is involved in the adjustment of nodule permeability to O2. Indeed the homologous gene in M. truncatula has been shown to restrict its expression in the cortex of mature functional nodule, and some cells of the inner-cortex of soybean nodule are hypothesized to be responsible of the variations in the cortex resistance to O2 entry inside this organ. In the present work is described a methodology of in situ hybridization with a radioactive probe on cuts of common bean nodules after embedding in a methacrylate resin. The results show an expression of the gene that is restricted to nodule inner-cortex, without any detectable expression in middle or outer cortex. It is concluded that the ponctual revelation of transcripts hybridization with this technique makes it conceivable to quantify hybridization spots in order to address whether their number in nodules from various environmental treatments, like phosphorous deficiency, does vary and correlate with variations of nodule permeability.
