Résumé : The purpose of this mission to the Coto laboratory from 23 to 28 June 2003 was to coordinate transferral to Coto of the different oil palm vegetative propagation protocols available in Montpellier. The difficulties encountered with implementing CIRAD protocol A on a solid medium are due to a technical problem with culture medium manufacture. The last two factors remaining to be checked are the Agar and the autoclaving temperature. Precise instructions on this aspect were provided and a sample of the Agar used in Montpellier was supplied. Once conditions for making the culture medium are satisfactory, all the cultures should be transferred to new media, for all stages of the procedure. The most spectacular results are for establishing cell lines in liquid medium. Forty-seven different clones are now being cultured using this procedure and it can be considered that the transfer of embryogenic oil palm lines to liquid medium is not a limiting factor for development of this procedure. A comparison of two culture medium sequences for obtaining embryos and shoots led to the protocol recommended by CIRAD (protocol B) being adopted, though bearing in mind that we do not have any hindsight on the use of cytokinins, and use of this compound needs to be validated with regard to the occurrence of phenotypic variants. Several technical improvements to the late development phases were considered and will be proposed at a later date. Lastly, a visit to the Montpellier team by Dr S. Jouannic for mature palm apex sampling has been prepared.
