Titre anglais : Diversity and diagnosis optimization of the phytoplasma associated to the coconut lethal yellowing in Cuba

Résumé : Between the aspects of greater interest in the international scope with respect to the Coconut Lethal Yellowing (LY) are the study of the genetic variability of phytoplasmas associated to the disease and the improvement of the methods for their diagnosis, those that constitute objectives of this work. For the optimization of the diagnosis, different primers pairs used for the detection of the disease in other Caribbean regions were tried (LY1F/LY1R and MMR/MMF), as well as distinct concentrations from MgCl2 and dilutions of the samples. Twenty-three positives isolated coming from five Cuban provinces were selected and they were analyzed by PCR-RFLP, using universal primers P1 and P6 (Deng and Hiruki, 1991) and restriction enzymes EcoRl, Rsal and BamHl. The concentration of 2.5mM Of MgCI2 and the sample dilution of 1/100 were optimal for the diagnosis with primers LY1F/LY1R. No amplification was obtained when MMR/MMF primers were used. Comparison between restriction patterns evidenced the diversity of Cuban isolates, the presence of 5-7 profiles and the main ones with each enzyme.
