Résumé : L'accident nucléaire ayant eu lieu à Fukushima le 11 mars 2011, a entrainé une libération de radionucléides, principalement du 137césium, retenu par les argiles dans les premiers centimètres du sol. L'objectif de la thèse a consisté à obtenir des idéotypes de riz limitant l'entrée du 137Cs par les racines, et sa translocation vers les parties aériennes. Afin de répondre à cette problématique, l'identification et la caractérisation des gènes impliqués par l'entrée du césium dans la plante a été une étape primordiale. Les résultats obtenus en levures mais également in planta, ont confirmé l'implication du transporteur potassique OsHAK1 comme étant le transporteur majeur. Un criblage de mutants de levures pour le gène OsHAK1 a permis de générer 2 mutations qui entrainent une réduction de la perméabilité du transporteur pour le Cs+. Une validation in planta à l'aide de la technologie Crispr/Cas9 permettrait d'insérer de façon spécifique les différentes mutations lors de la coupure double brin dans le gène OsHAK1, par recombinaison homologue. Le césium étant concentré uniquement dans les premiers centimètres du sol, l'architecture racinaire représente un enjeu majeur en termes d'absorption du césium. Ainsi, le second volet de l'étude consistait à évaluer l'influence de la modification de l'architecture racinaire du riz sur sa capacité à capturer le radio-élément dans les horizons superficiels du sol. Pour cela, nous avons fait appel à la diversité naturelle ou induite présente dans l'espèce Oryza sativa L. En plus de la lignée NIL-DRO1, connue pour son enracinement profond, nous avons fait le choix d'utiliser une lignée quasi isogénique affectée dans le gravitropisme de l'appareil racinaire. Nos résultats n'ont pas permis de confirmer statistiquement qu'un enracinement profond limite l'acquisition et le stockage du césium dans les parties aériennes, elles nous ont toutefois permis d'améliorer la caractérisation des matériels indica et japonica à travers un dispositif expérimental conçu pour mimer une contamination artificielle retrouvée à Fukushima. L'ensemble des résultats obtenus dans le cadre de cette recherche pourraient avoir des applications directes en termes de conduite à adopter en cas de fuite de radioactivité au niveau des centrales nucléaires localisées près de cultures agricoles.

Résumé (autre langue) : The nuclear accident in Fukushima on 11 March 2011 resulted in the release of radionuclides, mainly cesium, in the atmosphere, which contaminated the first few centimeters of soil where they remain retained by clays, notably in paddy fields. The objective of the thesis was to obtain ideotypes of rice (Oryza sativa L.) limiting the entry of 137Cs+ by the roots, and its translocation to the aerial parts. The first part consisted in identifying and characterizing genes involved in the entry of cesium into the plant. The results obtained in planta through reverse genetics showed the major involvement of the potassium transporter OsHAK1 in root Cs+ uptake from low concentrations. A screening in yeast of OsHAK1 mutants, obtained randomly by PCR, made it possible to identify 2 mutations which lead to a reduction in the permeability of the transporter to Cs+. An in-planta validation through a Crispr / Cas9 mediated specific insertion of the different mutations in OsHAK1 gene by homologous recombination has been undertaken. As cesium is only concentrated in the first few centimeters of the paddy field soil, the second part of this work consisted in evaluating the influence of the modification of root architecture on rice ability to capture the radio-element in the top soil. In that aim, we used the natural or induced allelic diversity existing in the DEEPER ROOTING 1 (DRO1) gene involved in deep rooting in rice. We have compared under controlled conditions a near isogenic line NIL DRO1 and a mutant dro1 line to their respective controls for their ability to accumulate cesium when grown onto a contaminated top paddy field soil. Though our results have so far not been able to statistically demonstrate that deep rooting limits cesium uptake and storage in aerial organs, they allowed the characterization of japonica and indica materials differing only through their root system distribution and the design of an experimental set up mimicking an artificial superficial contamination found in Fukushima paddy fields. All the results obtained in the course of this research could have direct applications in terms of the behavior to be adopted in the event of leakage of radioactivity at the level of nuclear power plants located near crops.
