2001. Identification of regulatory sequences in the 5' upstream region of hevein and rubber elongation factor gene from Hevea brasiliensis.

Résumé : The CaMV 35S promoter has been studied extensively as a source of DNA regulatory sequence to over-express genes in transgenic plants. It has also been shown that higher plant genes that encode messenger RNA contain distinct regulatory sequences in the 5¿flanking region that are required for tissue-specific statement. To construct a vector that is capable of directing statement of the protein in a specific tissue, a tissue-specific promoter is required. For example, to produce a desired expression in the seeds of transgenic plants, a seed-specific promoter is required. In CaMV35SGUS transformants of Hevea brasiliensis, the CaMV35S promoter facilitates strong GUS statement in the latex, although expression of the protein is still evident in other Hevea tissues. It may not be beneficial for transgenic Hevea to have recombinant proteins expressed unselectively in all its tissues as some of these proteins could be toxic or otherwise deleterious to the plant. Hence, it is desirable to isolate promoters which facilitate latex vessel-specific expression. One possible candidate promoter for latex vessel-specific expression is the hevein gene promoter. Hevein is among the most abundant proteins found in Hevea latex. The hevein controlling sequences, presumably associated with the promoter and 5¿ prime untranslated sequence of the hevein gene, direct high level expression in the latex tissue and target the protein to the lutoids (specialised organelles) in the latex. The hevein promoter could therefore be potentially used to facilitate latex vessel-specific expression. A study was thus initiated to isolate the regulatory sequences in the 5¿ upstream region of hevein.
