Titre anglais : Integrated control of bacterial wilt (Ralstonia solanacearum) of potato in the Vakinankaratra region

Résumé : Le flétrissement bactérien dû à la bactérie phytopathogène Ralstonia solanacearum est une maladie majeure de la pomme de terre dans la région Vakinankaratra, le principal bassin de production. Au cours de ces dernières années, cette maladie est à l'origine de baisses significatives du rendement et de la qualité de la production. Aucune méthode de lutte chimique n'est disponible et les mesures préventives préconisées s'appuient notamment sur le maintien ou le renforcement de la propriété de résistance de la plante hôte. Dans ce cadre, l'étude de la diversité des souches de R. solanacearum présentes dans les zones de production de la pomme de terre de cette région, prérequis pour toute stratégie de lutte génétique a été initiée. La potentialité des microorganismes rhizosphériques et endophytes des pommes de terre à limiter le développement de cette bactérie phytopathogène a également été explorée en vue de la mise au point d'agent de lutte biologique. En outre, l'impact de la diversité entomofaunique des plantations de pomme de terre dans la transmission de R. solanacearum a été évalué. La caractérisation moléculaire des souches a révélé que 735 isolats bactériens sont bien assignés à l'espèce Ralstonia solanacearum, dont 604 souches tombant dans le Phylotype II et 131 dans le phylotype III. Aucune souche de phylotype I n'a été décrite dans cette étude bien que leur existence soit signalée à Madagascar. L'étude plus poussée des caractéristiques biochimiques a montré que 24 isolats de R solanacearum obtenus à partir de 6 échantillons de sols rhizosphériques de pommes de terre malades appartiennent au biovar 2. Une collection de 77 souches a été constituée à partir de 6 échantillons de sols rhizoshériques et 6 échantillons de matériel végétal de pommes de terre saines dans le district d'Antsirabe II dont 40 Actinomycètes telluriques, 25 Actinomycètes endophytes et 12 Pseudomonas fluorescents. L'antagonisme in vitro des isolats d'Actinomycètes et de Pseudomonas vis-à-vis des souches de R. solanacearum a été mis en évidence par la technique de diffusion sur gélose. Parmi les 77 souches isolées, 12 ont présenté une activité inhibitrice à l'égard de souches de R. solanacearum testées. L'inoculation en serre de plant de pomme de terre avec les 4 souches sélectionnées a permis de réduire l'incidence de la maladie jusqu'à 19 % et d'augmenter l'efficacité du biocontrôle (plus de 70 %) pour la souche d'Actinomycète endophyte E7. Cette expérience a permis également d'obtenir une augmentation significative de plus de 45 % de la croissance des plantes. Par ailleurs, de larges groupes d'insectes évoluant dans les plantations de pommes de terre ont été observés dont les lépidoptères en tant que défoliateurs, des hémiptères en tant que piqueur-suceurs et de coléoptères comme foreurs. Les variétés de pomme de terre présentant les plus fortes incidences de la bactériose ainsi que le maximum de nombre de familles d'insectes, dont notamment les coléoptères sont Spunta, Bandy akama et Meva.

Résumé (autre langue) : Bacterial fading due to the bacterium phytopathogene Ralstonia solanacearum is a major disease of potato in the area Vakinankaratra, the principal coalfield. During these last years, this disease is at the origin of significant decreases of the output and the quality of the production. No method of chemical fight is available and the recommended preventive measures are based in particular on the maintenance or the reinforcement of the property of resistance of the plant host. Within this framework, the study of the diversity of the stocks of R. solanacearum present in the production zones of potato of this area, prerequisite for any genetic fight plan was initiated. The potentiality of the micro-organisms rhizospheric and endophytes of potatoes to limit the development of this bacterium phytopathogene was also explored for the clarification of agent of biological fight. Moreover, the impact of the entomofaunic diversity of the potato plantations in the transmission of R. solanacearum was evaluated. The molecular characterization of the stocks revealed that 735 bacterial isolates are well assigned with the species Ralstonia solanacearum, from including 604 stocks falling into Phylotype II and 131 in the phylotype III. No stock of phylotype I was described in this study although their existence is announced to Madagascar. More pushed study of the biochemical characteristics showed than 24 isolates of R. solanacearum obtained starting from 6 samples of sick potato grounds rhizospheric belong to the biovar 2. A collection of 77 stocks was made up starting from 6 samples of grounds rhizosheric and 6 samples of vegetable healthy potato equipment in the district of Antsirabe II including 40 Actinomycètes telluric, 25 Actinomycètes endophytes and 12 Pseudomonas fluorescent. The in vitro antagonism of the isolates of Actinomycètes and Pseudomonas with respect to the stocks of R. solanacearum was highlighted by the technique of diffusion on agar. Among the 77 isolated stocks, 12 presented an inhibiting activity with regard to stocks of R. solanacearum tested. The inoculation in greenhouse of potato seedling with the 4 selected stocks made it possible to reduce the incidence of the disease up to 19 % and to increase the effectiveness of the biocontrôle (more than 70 %) for the stock of Actinomycète endophyte E7. This experiment also made it possible to obtain a significant increase of more than 45 % of the growth of the plants. In addition, broad groups of insects evolving in the potato plantations were observed of which lepidopterans as défoliateurs, of the hémiptères as stitchernozzles and coleopters as borers. Varieties of potato presenting the strongest incidences of the bactériose like the maximum of number of families of insects, whose in particular coleopters are Spunta, Bandy akama and Meva.
